O'Donnell was born and raised in Commack, Long Island, New York, as the third of five children. Roseann Teresa (née Murtha; 1934–1973), a homemaker, and Edward Joseph O'Donnell (1933–2015), an electrical engineer who worked in the military industry, were her parents. [4] Her mother was Irish American, and Edward had immigrated from County Donegal, Ireland when he was a child. O'Donnell grew up as a devout Catholic. Daniel J. O'Donnell, her older brother, is now a member of the New York State Assembly. O'Donnell's mother died of breast cancer four days before her 11th birthday, on March 17, 1973.
From 1979 through 1984, O'Donnell worked as a stand-up comedian in clubs. On Larry King Live, she explained how she received her first major break on Star Search:
I was 20 years old and working at a Long Island comedy club. I was approached by a woman who remarked, "I think you're humorous." Could you please provide me your phone number? Ed McMahon is my father's name. Yeah, yeah, yeah, yeah, yeah, yeah, yeah, yeah, yeah, yeah, yeah, yeah My father's phone number was given to her. I was still living at home, so whatever. Three days later, the Star Search talent booker called and said, "We're going to fly you down to L.A." I've won five weeks in a row. It also provided me with national visibility.
