Lightfoot went on to work as an attorney at the Mayer Brown law firm, where he served a diverse range of clients. Lightfoot began her career as an Assistant United States Lawyer for the Northern District of Illinois. Police Superintendent Terry Hillard appointed Lightfoot as top administrator of the Chicago Police Department Office of Professional Standards, a now-defunct governmental police monitoring agency, in 2002. She was in charge for two years. Mayor Richard M. Daley appointed her as deputy chief of the Chicago Department of Procurement Services. In Illinois, Lightfoot has also served on the boards of the NARAL and ACLU chapters. Lightfoot was a contender for the office of United States Attorney for the Northern District of Illinois in 2013.
In 2015, Mayor Rahm Emanuel chose Lightfoot to succeed 19-year incumbent Demetrius Carney as head of the Chicago Police Board. In the aftermath of the Laquan McDonald murder issue, Emanuel also appointed Lightfoot to lead a special Police Accountability Task Group. Lightfoot was reappointed as head of the Police Board by Emanuel in 2017.
Lightfoot retired from the Police Board in May 2018, just before launching her candidature for mayor. Lightfoot launched her campaign for Mayor of Chicago in the 2019 elections on May 10, 2018, her first attempt for public office. She is Chicago's first Gay mayor and the city's first black female mayor. In the 2019 Chicago mayoral election, Lightfoot campaigned for mayor, reaching a runoff against Toni Preckwinkle, which Lightfoot won on April 2, 2019. In the initial phase of the 2023 Chicago mayoral election, she was defeated for re-election.
